The Agilent 8453 is a diode array UV-Visible spectrophotometer that measures the absorption of ultraviolet and visible light by liquid samples. This instrument operates across a wavelength range of 190-1100 nm with a wavelength accuracy of < ± 0.5 nm and measurement reproducibility of < ± 0.02 nm, enabling rapid full-spectrum acquisition in under one second. Samples are typically loaded as aqueous or organic solutions in 1 cm quartz or plastic cuvettes, though other configurations are possible. A sample thermostat is available for temperature-controlled measurements ranging from -20 to 110°C.
The system is commonly used for quantitative analysis of analyte concentrations, kinetic studies of chemical reactions, and qualitative identification of compounds based on their characteristic absorption spectra. Applications include measuring protein and nucleic acid concentrations, determining chemical purity, analyzing dyes and pigments, and studying chromophore-containing molecules.
